Pegula was given a wildcard into the main draw of the US Open doubles tournament on August 30, 2011, where she was paired with Taylor Townsend. They eventually lost in the third round to Vania King and Yaroslava Shvedova, the third-seeded team. Pegula was given a wildcard to the qualifying draw in Indian Wells in March 2012, and she surprised higher-ranked players Bojana Jovanovski and Paula Ormaechea to qualify for the main draw, where she lost in three sets to Magdaléna Rybáriková.
Pegula made her Grand Slam singles debut as a qualifier at the 2015 US Open. She defeated Shuko Aoyama, Margarita Gasparyan, and Melanie Oudin on her way to the main draw, where she was defeated in the first round by Alison Van Uytvanck.
